Bacterial community DNA metabarcoding
UNDER EMBARGO - This dataset is part of BE/2023 sampling campagn in SW Greenland fjords (Igaliku and Tunulliarfik). Samples for DNA extraction were collected along fjord transects at several depths of the water column. The focus of the study was to determine the taxonomical composotion of bacterial community in two Arctic fjords.

- Seawater DNA was amplified targeting the full-length 16S rRNA gene using primers 27F and 1492R (Lane, 1991). PCR, library preparation, and barcoding followed Van Der Loos et al. (2021), including mock community and triplicate samples for quality control. Amplicons were sequenced on an Oxford Nanopore MinION platform (R9.4.1 flow cell, 72 h run). Raw reads were basecalled with Guppy, quality-checked (NanoPlot), and demultiplexed (qcat). Chimeras were removed (Yacrd), and reads were filtered by length (1000–2000 bp) and quality (Q >10) using NanoFilt. Sequences were clustered into Operational Taxonomic Units (OTUs) at 97% similarity, and taxonomy was assigned using Kraken2. Downstream processing (phyloseq) included removal of singletons, doubletons, non-bacterial sequences (chloroplasts and mitochondria), unclassified OTUs at phylum level, and potential contaminants (>1% relative abundance in negative controls).
